Some skilled occupations - notably those that are assessed by the assessing body called VETASSESS, but also teaching, accounting, and healthcare occupations - require you to have qualifications to secure the skills assessment classification which is a requirement before you submit the visa application to the Department of Immigration. Other occupations allow a positive skills classification based on work experience alone - eg trade skills occupations assessed by Trades Recognition Australia. Best regards.
